The 6 button layout is good for most Capcom fighters. But, Marvel vs Capcom 2 has a redesigned button layout designed to be easier to use. Also the new layout allows you to call your helper characters. The layout looks like this:
1(LP) 3(HP) (Assist 1)
2(LK) 4(HK) (Assist 2)
So what happened to the medium punch and medium kick? You can still get them by pressing LP twice in a combo. The magic series would look like this: LP, LK, LP, LK, HP, HK. Lets break this down.
This is what you press:
LP LK LP LK HP HK
This is what your character does:
LP LK MP MK HP HK
So what happens to people with MP or MK as launchers? You have to press LP, MP to launch them. As an alternative, you can just press DF + HP and it will do MP launchers for people like Cyclops without having to combo the LP into it. Capcom thought of everything.
Lets talk about the Assist buttons and their uses. In MVC2 (Marvel VS Capcom 2 abreviation. Get used to seeing it written that way.) when your character's life bars are arranged in this order: Point character (the fighter thats on screen fighting), assist 1 aka the second character, assist 2 aka the third character. When you press assist one, the second fighter will come out and do a quck attack, taunt, then leave the screen. If you press assist 2, the third fighter will come out and do a quick attack, taunt, then leave the screen. Press them together to get a Team hyper combo when you have 3 levels of super meter. If you only have one level, then only your point character will do his or her super.
In MSHvsSF and MVC1, you can call your assist by pressing Mp + Mk. In MSHvsSF, your partner comes out and does a quick attack, taunts and leaves. In MVC1, you pick your assist character at the begining of each round. You have a limited number of uses for that assist (they can only come out around 5 times per match) and you cant actually fight with them like you can in MSHvsSF and MVC2.
